Private Schools vs. Public Schools Sports Teams - 1606 Words

The imbalance between public and private school sports teams does not need to be adjusted. Private schools are offering an extra service that public schools cannot offer. That service is better teachers all the way to better coaches. There are no rules on how they can raise money for their athletic programs unlike public schools. Because of the athletic success more student athletes want to go to the private schools. Which in turn gives the student athletic a better opportunity to be recruited. No the private schools do not need to be adjusted public schools need to improve their outlook on athletics. (Popke, 2006) With this system in place they feel that this the only way to level the playing field. Another state Tennessee has used the multiplier policy and a separate division for all private and non-public schools that offer financial aid to the varsity athletes. For the nonpublic schools that do not render financial aid can still can compete in division I but to make the playing field even they must multiply the enrollment by 1.8 which moves them up a class. (Popke, 2012) Programs recruit themselves When you look at the success of private schools in athletics there is no need to recruit students. Just like a public magnet high school students come there because of the special services they offer. This is where the public schools need to adapt their way of thinking. The question is why students are leaving public schools to attend private schools. Lets start with coaches at a public high school. With the budget cuts more coaches are leaving public high schools this makes it hard for athletic directors to hire the right coach. I answer for the lieutenant’s loyalty to the Foundation. â€Å"Now I was in touch with him the day after we stopped getting the daily report from Munn-â€Å" â€Å"Why?† broke in Darell, fiercely. â€Å"I thought it was quite decided that we were not to make a move in the matter. You were risking their lives and ours.† â€Å"Because,† was the equally fierce retort, â€Å"I’ve been involved in this game for longer than you. Because I know of certain contacts on Kalgan of which you know nothing. Because I act from deeper knowledge, do you understand?† â€Å"I think you’re completely mad.† â€Å"Will you listen?† A pause, and Darell’s eyes dropped. Anthor’s lips quirked into a half smile, â€Å"All right, Doc. Give me a few minutes. Tell him, Dirige.† Dirige spoke easily: â€Å"As far as I know, Dr. Darell, your daughter is at Trantor. At least, she had a ticket to Trantor at the Eastern Spaceport. She was with a Trading Representative from that planet who claimed she was his niece. Your daughter seems to have a queer collection of relatives, doctor. That was the second uncle she had in a period of two weeks, eh? The Trantorian even tried to bribe me – probably thinks that’s why they got away.† He smiled grimly at the thought. â€Å"How was she?† â€Å"Unharmed, as far as I could see. Frightened. I don’t blame her for that. The whole department was after her. I still don’t know why.† Darell drew a breath for what seemed the first time in several minutes. He was conscious of the trembling of his hands and controlled them with an effort. â€Å"Then she’s all right. This Trading Representative, who was he? Go back to him. What part does he play in it?† â€Å"I don’t know. Do you know anything about Trantor?† â€Å"I lived there once.† â€Å"It’s an agricultural world, now. Exports animal fodder and grains, mostly. By evening, the exteriormost reaches of Dr. Darell’s personality, the ones that made immediate contact with other people had solidified once more. He had refused to eat his evening meal and had, instead, with feverish insistence, returned to the inchwise advance into the intricate mathematics of encephalographic analysis. It was not till nearly midnight, that he entered the living room again. Pelleas Anthor was still there, twiddling at the controls of the video. The footsteps behind him caused him to glance over his shoulder. â€Å"Hi. Aren’t you in bed yet? I’ve been spending hours on the video, trying to get something other than bulletins. It seems the F.S. Buddhism: Things I Find Interesting As I was reading the selected portions of the book for this chapter, I came across a few things that I found interesting. At first I did not catch them, but after I went back and reread the selections, I found these things, that I thought were intriguing. Buddhism is supposedly a non-theistic religion. However, in the reading titled "The Majjhim-Nikaya: Questions Which Lend Not to Edification" (5.1) and in "Realizing the Four Noble Truths" (5.3, the Buddha is continually referred to as "The Blessed One". If he is only a man, why is he referred to in such a way? Other Buddhists have the potential to become Buddhas, so would they also be referred to as "Blessed One"? If this Buddha was not the first, and not the last Buddha, why are other Buddhas also not referred to as "Blessed ones"? In my opinion, if this Buddha is not considered to be a god, then he should at least be referred to as "One of the Blessed Ones" instead of "The Blessed One". Another concept that I found interesting was the idea of duality that was discussed in "The Majjhima-Nikaya: Questions Which Lend Not to Edification" (5.1). I do not quite understand it, so I was intrigued it. The selection says that the soul and body are identical, and then that the soul and body are separate. It says that the world is finite and that the world is infinite. It also says that the world is eternal and then that it is not. How can these things be? I do not understand how the saint can both exist and not exist after death. I think that in my worldview, things such as these concepts are not present. I see ideas such as the world, and the body and soul as one way or the other.

Shortly after being employed Dick secured a single, large contract deal, placing him high on the sales leader board. Dick was confident in his abilities and took pride in getting the job done. It’s important to have confidence at work to professionally grow and advance your career. Dick’s confidence as a salesman later helped him get a management position. Dick enjoyed being a salesman and was pretty good at it. They also reported improvements in employees completing their work duties and a willingness to make decisions (Gyllensten and Palmer, 2014). Again we can also give credit to Dick’s MBA degree as providing self-assurance in his job performance abilities (Simpson, 2005). Confidence gives you the courage to do things you normally wouldn’t feel comfortable doing. In sales there are plenty of circumstances where the ordinary sales pitch just won’t seal the deal. Dick was also very committed to the job which reflected in his sales volume. In a review of the research on organization commitment and job performance, researchers identified there is a direct correlation in commitment to the job and achieving favorable results (Myer, Paunonen, Gellatly, Goffin, Jackson, 1989). Consistent with this prospective, Shore and M artin (1989) found that â€Å"better performance was shown by employees with higher commitments.† Dick was so committed to the job that it cost him his first marriage. He was serious about reaching his sales goals and each of these factors previously described played a part in his success as a salesman. However after a few years as a salesman, Dick wasn’t getting the satisfaction from the job as he had in the past. Therefore he requested a transfer out of the sales department and into a management role. Dick was named plant manager at Modrow. He came in at a hectic time when a lot of changes were being implemented. In his efforts to familiarize himself with the daily operations, Dick first struggle as a manager was micro-managing. He had his hand in everything possible which made his employees feel very un-easy. According to Dr. White, Dick possessed many symptoms of a Micromanager. For example, Dick interest in cutting costs required the expert knowledge of the accounting group. Knowing that  accounting wasn’t his strongest subject he still spent numerous hours burying himself in details that he just could not understand. Micromanagers don’t allow any decisions be made with their approval (White, 2010). Dick had also taken on too many tasks at once making it almost impossible for any of them to be implemented successfully. Being the micromanager that he was, Dick didn’t delegate tasks very well, another symptom of a micromanager (White, 2010). Another example of Dick’s micromanaging is when he insisted on changing the disposal process of scraps. When discussing this change with his foremen, Dick didn’t listen to the foreman’s thoughts as to why the change wouldn’t work. Micromanagers tend to have the mentality; it’s my way or the highway. Traditional micromanagers will completely disregard new concepts offered by their subordinates (Weyand, 1996). Work-related stress affected Dick’s job performance. It also was creating issues with his family as they felt abandoned while he spent countless hours at the office. Through trying to prove himself to leadership and to his new employees, Dick lost focus on other important matters. His management position required him to take on more responsibilities which demanded more of his time. â€Å"In order for supervisors and managers to excel and be a dynamic force in organizational life, job stress must be harnessed† (Stanley, 2014). As Dick noticed processes that needed to be changed he was unsuccessful in communicating those changes. As a manager being able to successfully communication is essential. Communication failure could potentially become a serious problem in an organization. Information is lost causing confusion reducing productivity and increases costs. In an article published by Jost (2006), he discusses several situations where failure to communicate resulted in some hefty financial costs to many companies. Dick was so focused on cutting costs that he forgot to look at the big picture. If he had effectively communicated his interest to cut costs with his supervisors and foreman, he could have potentially found a solution. Additionally, his relationships with his employees weren’t the best, which is another consequence of ineffective communication (Tunk, 2014). Now I’d like to focus on the following recommendations that could resolve Dick’s  micro-managing style, work-related s tress, and ineffective communication. To address Dick’s micro-managing style, he needs to be flexible in how the job gets done. Dick can delegate tasks that can be handled without his supervision and give them ample time to complete each task. He can do this by building trust, empowering his employees to be a part of the decision making process (White, 2010). Making rounds within the plant had a negative impact on employees. They were worried about what he was up to and why he kept coming around. There was a lack of trust. His presence caused everyone to be less productive in their jobs. The second recommendation is time management to address Dick’s work-related stress. It’s important for Dick to find a balance between work and family for the sake of his marriage and kids but also for his well-being. If Dick continues operating this way it could potentially lead to some serious health problems (Heikkia et al., 2013). To help him find this balance he needs to reevaluate what’s most important to him. Review his job duties that need to be completed and prioritize them. Identify the tasks that can be done by someone else and delegate those tasks (McDonald & Hucheson, 1998). Dick should also try to engage in outside activity that requires his complete attention. Research supports â€Å"that a psychological detachment from work effectively mitigates some of the negative effects of work-family conflict† (Moreno et al., 2009). My last recommendation is improving his communication skills. Dick was not successful in trying to implement a change in the process to discard scrap siding. The keys to effective communication is listening and learning. Effective communication improves work performance and productivity within the business. It also builds trust and creates transparency which is something Dick is currently lacking within the Modrow plant. Communication promotes collaborative work where everyone has an opportunity to share information (Tunk, 2014). Chinese clothing is an important part of their culture. Although China no longer dresses in their older more traditional styles, the traditional garments are still worn for holidays and ceremonies. There have been many historical changes in Chinese clothing, and the Chinese style choices vary depending on what region is being scrutinized. Hanfu is a term that includes all traditional Chinese dress worn prior to Manchurian and western influences. All Chinese citizens living under the Ming dynasty, and many dynasties prior to it adopted this style of dress. It was widely worn by all until the beginning of the seventeenth century and the start of the Quing dynasty. Hanfu, which was the widely accepted style in China for over three thousand years, consists of the Shenyi. A one-piece robe, which wraps around the body, the Shenyi is made up of different parts. The Jin, or the upper front of the body, the Jiaoling Youren, which means â€Å"crossed collars to the right† in Chinese and is the collar usually seen on the Hanfu of both men and women. The collar is only tied to the left as burial dress. The Qu and Mei are the cuffs and sleeves, and the Chang is the name of the skirt. The first recorded dynasty in China was the Shang dynasty. Although they did wear Hanfu clothing, most specifically the Shenyi, the garment worn by them was actually made of two separate pieces. The jacket, referred to as the Yi, and the skirt, or the Shang. It did not become a one-piece garment for many years. During the Shang dynasty the colors most often used for the garments were warm colors such as red and yellow. As time moved forward the Chinese began to decorate the fabrics with red and yellow designs. Different regions of the country had different norms as far as Hanfu dress. During the Zhou dynasty the western people used different styles of the Hanfu Shangyi to differentiate between classes. The differences in classes are shown through the two styles of sleeves, the broad and the narrow, and the decorations that the wearer hung from his or her belt such as pieces of jade. When the Quing dynasty took over, so did a new style of clothing. The Qipao was the traditional style for women and the Cheongsam was the style commonly worn by men. All Chinese were required to switch from the old Hanfu style of dressing or they would be killed due to the new rulers in place. The Qipao is a one-piece dress that traditionally fits loosely on the body and was designed to hide the woman’s figure. It is also sometimes referred to as the mandarin gown. In the early nineteen hundred the Qipao was updated into the dress we recognize as traditional Chinese dress today. The close fitting dress with a high neckline is now often worn as a party dress and has been copied and worn by American women as well. The Cheongsam is the men’s version of the Qipao. It is a two-piece outfit made up of a pair of simple pants and a top with a high neckline extremely similar to that of the Qipao. The Cheongsam is traditionally made in the color black, and was the formal dress worn by Chinese men up until the introduction of the western-style suit.

The Polymerase Chain Reaction Pcr The polymerase chain reaction was first developed in 1983 by Kary Mullis. This reaction is commonly used in molecular biology to amplify and generate thousands to millions of copies of specific DNA sequences across several orders of magnitude (4-1). It relies on thermal cycling, consisting of cycles of denaturation, primer (short DNA fragment) annealing and primer extension (4-7). PCR can also be used for the analysis of RNA sequences and to qualitatively detect RNA expression levels through creation of complementary DNA (cDNA) transcripts from RNA by use of reverse transcriptase. This technique is called reverse transcription-PCR (RT-PCR) (5-2). Although PCR and RT-PCR have revolutionized many areas of biomedical science, they are not suitable for the quantitative analysis of analysis of samples. Hence, real-time or quantitative PCR (qPCR) techniques need to be employed (5-8, 5-9). RT- qPCR distinguishes itself from other methods available for gene expression, such as northern-blot analysis, ribonuclease (RNase) protection assay and competitive RT-PCR, in term of accuracy, sensitivity and fast results (2,6). RT-qPCR does not required post-amplification manipulation and it can produce quantitative data with wide dynamic range of detection (7 to 8 logs). In addition, RT-qPCR assay is 10,000 to 100,000-fold more sensitive than RNase protection assays and 1000-fold more sensitive than dot blot hybridization (3). RT- qPCR also can even detect a single copy of a specific transcript and can reliably detect gene expression differences as small as 23% between samples (3-6, 3-7). Furthermore, it has lower coefficients variation (cv; TaqMan at 24%; SYBR Green at 14.2%) than end point assays such as probe hybridization and band densitometry (45.1%; 44.9% respectively) (3-8). RT- qPCR can differentiate between messenger RNAs (mRNAs) with almost identical sequences and requires much less RNA template than other methods of gene expression analysis. Because of this, RT- qPCR has established itself as the gold standard for the detection and quantification of RNA targets (1-2,2). Furthermore, it is firmly established as a mainstream research technology (1-3). However, the major disadvantage of RT-qPCR is that required expensive equipment and reagents (3). The principle of RT-qPCR is straight forward: following the reverse transcription of RNA in to cDNA, it needs an appropriate detection chemistry to dete ct the presence of PCR products, an instrument to monitor the amplification in real-time and compatible software for quantitative analysis. RT- qPCR is characterized by the point in time during cycling when a PCR product amplification is first detected (Figure 1, 1). A direct relationship between the starting copy number of the nucleic acid target and the time required to observe fluorosence increasing. Nowadays, there are four fluorescent DNA probes available for RT-qPCR detection of PCR products: TaqMan, SYBR Green, Molecular Beacons, and Scorpions. All of them generate a florescent signal to allow the detection of PCR products. While the TaqMan probes, SYBR Green, Molecular Beacons, and Scorpions generation of fluorescence depend on Forster Resonance Energy Transfer (FRET) coupling of the dye molecule and a quencher moiety to the oligonucleotide substrates, the SYBR Green dye simply emits its fluorescent signal by binding to the double-strand DNA in solution (5-34). As RT-qPCR has extremely high sensitivity and reproducibility, in depth understanding of normalization techniques is imperative for accurate conclusions (6). Normalization of gene expression data is an essential component of a reliable RT-qPCR assay and it is used to control for error between samples (7,3). This error could be introduced at one or more stages throughout the experimental protocol; (input sample, RNA extraction, etc.) however, there are many strategies to control this error ( please, read the discussion section strategies for more details). Currently, internal control genes, which are often referred to as housekeeping genes, are most frequently used to normalize the messenger RNA (mRNA) fraction. This housekeeping gene should remain constant in the tissues on cells under investigation, or in response to experimental treatment (8, 8-69, 8-70, 6,7). In addition, the ideal housekeeping genes should by stably expressed, and their abundances should show strong correlation w ith mRNA total amounts present in the samples (8). Consequently, normalization against a single housekeeping gene in not acceptable and can falsely bias results unless the researchers present clear evidence for the reviewers that confirms its invariant expressions conditions described (3,8-71,8-73). In this study we carried out an evaluation the gene expression of three commonly used housekeeping genes (GAPDH, ÃŽÂ ²-action, ALAS1) in three different cell lines which are derived from T-cell leukemia, B-cell lymphoma and myeloid leukemia, using RT-qPCR as an analytical tool. Our goal was to recognize a housekeeping gene with minimal variability under different experimental conditions. Materials and Methods: Samples, RNA handling and isolation: Cell line pellets (5-10X106 cells) which have been frozen in 0.5 ml TRIsure Reagent (Bioline code BIO-38033). Cell lines are: Jurkat, CEM-C7 and MOLT-4 (all T-cell leukemia-derived); SKW 6.4, BJAB and JeKo-1 (all B-cell lymphoma-derived); and HL-60, NB4 and K562 (all derived from myeloid leukemia). RNA was isolated from cell pellets using Trizol procedure. However, isolating and handling RNA ask for special precautions because naked RNA is highly susceptible to degradation by ribonucleases (RNases) (1). RNases are found everywhere and they are very stable and active enzymes that do not require metal ion co-factors to function and can maintain activity even after prolonged autoclaving or boiling. So that, all equipment and reagents should be treated to inactive RNases prior to use. Wearing gloves while handling reagents and RNA samples, changing gloves frequently, keeping tubes closed whenever possible and keeping isolated RNA on ice when aliquots are pipetted for downstream applications could prevent RNase contamination. We are used sterile, disposable plastic ware and they were RNase-free and do not require pretreatment to inactive RNAses (8-46,8-47). The quantity of the isolated RNA was determined by nanodrop spectrophotometry (absorbance at 260 nm of a 40ÂÂ µg/ml soluti on of RNA is 1.0) using nuclease-free water as a blank. A sample was reserved for quality assurance (see below) and the remainder was stored at -80 degree centigrade for one week. Gel electrophoresis (quality assurance of RNA) : The quality of the isolated RNA was verified by agarose gel electrophoresis. The gel was run at 100V for 30 minutes and photograph under UV transillumination. DNA digestion: A DNase digestion step was performed as a precaution using RQ1 RNase-free DNase kit although the TRIzol method generally results in RNA which is essentially free from genomic DNA (2). A sample was reserved for reverse transcription (see below) and the remainder was stored at -80 degree centigrade. Reverse transcription: 11 ÂÂ µL of DNase-treated sample was reverse transcripted, using Superscript II reverse transcriptase, to complementary (cDNA) by random hexamer priming.
